A 30-year-old man received a broken leg and a 32-year-old man received superficial injuries.
It is believed the group had been involved in an altercation with another group of men close to John Dalton Street before the incident.
Superintendent Stuart Ellison from North Manchester Division said: “From what we know, a silver Corsa has driven at speed, on the wrong side of the road and collided with the three men.
“At this moment in time, we cannot rule out this was a deliberate act and we are keen for anyone who may have witnessed the collision to get in touch. A man is in a critical condition in hospital and two others have been injured and we need information that can help us find who is responsible.
“If you saw a silver Vauxhall Corsa speeding away from the John Dalton Street area in the early hours of this morning, please contact us. We believe the car turned left onto Deansgate and drove towards the south side of the city.
“Any information you have, no matter how insignificant you may think it is, could be vital for this investigation and we are keen to hear from you.”
